SATAN WEDNESDAY (Century of Sauton, the fr. bacteriologist, 19 — 20 centuries) — a fluid synthetic medium. It is offered by Soton in 1912 for cultivation of mycobacteria of tuberculosis. In further S. found for page application for cultivation of all types of mycobacteria, except mycobacteria of a leprosy (see. Mycobacteria ).
It is widely used at production of a vaccine BTSZH (see) and cleared tuberculine (see). The village of the village, being standard, does not contain foreign impurity, has considerable advantages before Wednesdays with organic components.
(On 1000 ml) are S.'s part of page: L-asparagine (4 g), glycerin (60 ml), lemon to - that (2 g), potassium phosphate dibasic (0,5 g), epsom salt (0,5 g), ammoniac ferric citrate (0,05 g), a distilled water.
For S.'s preparation by the village mix of its components is warmed up before full dissolution of salts. The cooled solution is alkalinized ammonia to pH 7,2, spilled in glass tanks and sterilized in the autoclave of 20 min. at t ° 120 °. After sterilization S. of page is transparent, slightly greenish shade, pH 6,8 — 7,0. On early propagation steps of mycobacteria of pH raises to 7,8 — 7,9 owing to eliminating of ammonia from asparagine, and then goes down due to oxidation of glycerin and reduction of the ions of ammonium used by a microbic cell for its growth.
Cultures of mycobacteria usually to — to the 8th day of cultivation form a solid dry wrinkled film, growth on S.'s surface of page the cut in thickness proceeds to 3 weeks. Increasing amount of potassium phosphate and adding microdose of zinc, it is possible to improve growth properties C. of page. At cultivation of a vaccinal strain of BTsZh for lack of zinc the film in the course of an incubation is humidified and sinks, an exit of microbic weight sharply decreases (to 20% in comparison with Wednesday, zincilerous). Also metabolism of a cell is broken, its antigenic structure changes — the maintenance of lipids and phospholipids increases a little and the content of hexoses and a protein decreases.
More than 20 modifications of S. of the page differing in nek-ry changes of composition of salt in solution of the environment and administration of microdoses of zinc are offered.
