HLORIDYN (Chloridinum; synonym: Daraclor, Daraprim, Malocide, Pyri-methaminum, Tindurin) — chemotherapeutic means for treatment of malaria and a toxoplasmosis. 2,4-Diamino-5-ya-chlorophenyl - 6-ethyls-pirimi-dynes; C12H13C1N4:

Inodorous white crystal powder and taste. Let's not dissolve in water, it is a little alcohol-soluble, non-hygroscopic.

Inhibits the digidrofolatredukta-storage and in this regard exerts the oppressing impact on development of causative agents of malaria and a toxoplasmosis. Activity of Chloridinum considerably increases at its simultaneous use with streptocides (see. Sulfanamide drugs). After intake it is well soaked up from went. - kish. a path and in the maximum concentration collects in blood in 2 — 6 hours. Apprx. 80% of Chloridinum connects

sya with proteins of plasma. The period of semi-removal makes 80 — 100 hour. It is allocated in not changed type of hl. obr. kidneys, partially sialadens, and in the period of a lactation and mammary glands. After a single dose circulates in blood in the concentration sufficient for suppression of erythrocyte forms of causative agents of malaria, during 1 week.

Chloridinum possesses gematoshizo-tropny (shizontotsidny) action and causes death of sexless erythrocyte forms of all types of plasmodiums. Exerts the oppressing impact on development of preeritrotsitarny schizonts, Plasmodium falciparum is preferential. Does not work on it is long the existing fabric forms (gipnozoita) of R. vivax and P. ovale. Has gamostatichesky (sporonto-tsidny) effect on sexual forms.

Chloridinum is applied generally to chemoprophylaxis of a tetrian fever (see). At acute disease it is usually not used because of rather slow approach of effect. Resistance to Chloridinum is eurysynusic at P. falciparum in Africa. It does not affect with per se the strains of P. falciparum steady against Chingaminum (see) that is often combined with polyresistance to the majority of antimalarial drugs. The effect at the tropical malaria caused by such strains can be reached at use of Chloridinum in a combination with streptocides or sulphones thanks to potentiation of its action.

Drug is used inside. For chemoprophylaxis of a tetrian fever Chloridinum is appointed the adult in a dose of 0,025 g once a week; to children — depending on age: till 1 year of 0,00625 g, 1 — 4 years — 0,00625 — 0,0125 g, 5 — 8 years 0,0125 g, 9 — 12 years 0,0125 — 0,025 g. Reception of Chloridinum is begun for 1 week or several days before danger of infection with malaria and continued during the entire period of possible infection, and then during 4 — 6 weeks after the end of this period.

At drug resistant tropical malaria Chloridinum is used in a combination with streptocides, hl. obr. the prolonged action (e.g., sulfalene), to-rye accept along with Chloridinum. At the same time Chloridinum appoint the adult on 0,025 g, children up to 4 years on 0,00625 g, 4 — 8 years on 0,0125 g, 9 — 14 years on 0,0187 g a day, and sulfalene — the adult on 0,5 g and children depending on age respectively on 0,125; 0,25; 0,375 g. Treatment of malaria carry out by the specified combination within 3 days


in a row in combination with quinine (see). For chemoprophylaxis this combination is appointed weekly according to the same scheme, as at prevention of a tetrian fever.

Chloridinum is applied also to treatment of a toxoplasmosis (see).

Side effect of Chloridinum is sometimes shown by headaches, dizzinesses, heartaches, dispeptic frustration, decrease in level of hemoglobin in blood. After drug withdrawal these phenomena quickly pass. Prolonged use of Chloridinum can be followed by the toxic manifestations caused by deficit folic to - you (megalobla-stichesky anemia, is more rare a leukopenia) in this connection for prevention and treatment of these manifestations use folic acid (see).

Contraindications to purpose of Chloridinum are diseases of the hemopoietic bodies and kidneys. Drug appoint with care at deficit folic to - you. In the first 9 weeks of pregnancy Chloridinum is not applied in order to avoid toxic action on a fruit. In later durations of gestation Chloridinum is used with care.
