BETA NAPHTHYLAMINE — the substance possessing cancerogenic action; arylamine of a naphthalenic row (2 naphthylamine, 2 aminonaphthalene):
Colourless or inodorous reddish plates; t ° pl 111 — 112 °; t ° kip 306,1 °. It is well dissolved in alcohol, ether, hot water, it is bad — in a cold water (1,17%); letuch with water vapor, sublimates.
Till 1950 B. - the N was applied to receiving some azoic dyes.
In 1938 in experience on dogs possibility of tumors of a bladder at daily introduction of B. is for the first time proved - N with food in number from 100 to 450 mg. In 20 months tumors of a bladder were found in 13 of 16 experienced animals. The same new growths were received at hypodermic introduction of B. - N. Further researches confirmed high carcinogenic activity of B. - N. It is established that at introduction of high doses of B. - N are found in dogs multiple tumors of a bladder already on 9 — 10th month.
It is considered that carcinogenic activity not B. - N, and his metabolites has: 2-amino-1-naphthol and 2-naftilgidroksilamin. At removal from an organism they collect in a bladder and, affecting his mucous membrane, lead to formation of tumors. It was established by researches on mice. To them entered into a bladder the parafino-wax pills containing B. - N or 2-amino-1-naftolgidrat, allocated from urine of dogs, the Crimea to food was added by B. - N. Tumors in a bladder were got only at those mice, the Crimea was entered by pills with 2-amino-1-naftolgidratom.
B. - N, having high carcinogenic activity, can cause at working with it tumors of a bladder (cancer and papillomas). According to the published data of the world literature, till 1962 more than 400 diseases of the professional tumors of a bladder caused by contact with B. - N are revealed.
B.'s production - N, existing in the USSR from 1930 to 1951 (with a break in 5 years), but to the requirement a dignity. bodies it was stopped. Some tens of kilograms a year of chemically pure B. - N are developed as a reactant. Technical B.'s production - N is stopped for hygienic reasons also in the majority of the countries of the world.
Some semi-products of the anilinocolourful industry which as impurity contain B. - N can constitute a certain danger, however: alpha naphthylamine, aldolyalfa-naphthylamine and aminotobias to - that.
Alpha naphthylamine (1 naphthylamine, 1 aminonaphthalene, azoamine of grenades C) receive from alpha nitronaphthalene recovery by a disulfide of sodium.
Impurity B. - N in technical alpha naphthylamine is caused by existence in initial technical alpha nitronaphthalene of impurity of beta nitronaphthalene which at recovery passes into B. - N. The technical alpha naphthylamine produced until recently in the USSR contains up to 4 — 5% of B. - N. The alpha naphthylamine produced in other countries also contains B. - N.
According to the Soviet and foreign authors, chemically pure alpha naphthylamine in itself has no cancerogenic properties, and its cancerogenic action is caused by B. which is contained in it - N. Preventive actions in these productions are similar to those which are carried out during the work with benzidine (see. Benzidine ). Dekantserogenization who is let out in the USSR alpha naphthyl-amine it is conducted in two directions: purification of technical alpha nitronaphthalene of beta nitronaphthalene and receiving alpha naphthylamine by catalytic interaction of other raw materials.
Aldolyalfa-naftilamin contains up to 15% of technical alpha naphthylamine and may contain up to 0,75% of B. - N.
With a dekantserogenization of alpha naphthylamine danger of cancerogenic influence for working as in production aldol - alpha naphthyl of amine disappears, and at its use in rubber industry.
Aminotobias acid technical contains 0,5% of B. - N.
It is applied in production of varnishes and varnish dyes. Experiments on dogs and rats showed that chemically pure aminotobias to - that has no cancerogenic properties, and the technical product has weak carcinogenic activity. Events for reduction of content of impurity B. - N in aminotobias to - those are held.
