BESTA METHOD (F. Best, 1878 — 1920, is mute. the doctor) — identification of a glycogen in microscopic sections of the fabrics which are previously concluded in photoxylin or paraffin.

We will well dissolve a glycogen in water and quickly breaks up after death (glycogenolysis) therefore both in the course of fixing of material, and at after-treatment of cuts it is better to avoid use of aqueous solutions, however some specialists consider danger of use of aqueous solutions of fixers exaggerated. As fixers use mix of 96% of alcohol with formalin (3: 1), liquid Carnoy (absolute alcohol 6 of h, chloroform 3 of h, ice acetic to - you are 1 h), mix of 96% of alcohol with picric to - that, etc.

Preparation of reactants: 1) Best's dye — 2 g of a carmine, 1 g of potassium carbonate and 5 g of potassium chloride dissolve in 60 ml of a distilled water and boil within 5 min., after cooling and filtering add 20 ml of ammonia (stock solution is suitable within 3 months at t ° 0 — 4 °); before the use add 15 ml of ammonia and 12,5 ml of methanol to 15 ml of stock solution (working solution is good 2 — 3 weeks); 2) Best's solution — absolute alcohol 8 of h, methanol 4 of h, a distilled water of 10 h.

Coloring of cuts: cuts will see on the battery of alcohols of the increasing concentration to absolute alcohol, then paint Ehrlich's hematoxylin (5 min.), rinse in alcohol and water, paint Best's dye (15 — 30 min.), differentiate in Best's solution, then wash out in alcohol, a xylol, conclude in balm. As a result of coloring of drug the glycogen becomes bright red, kernels of cells — blue. Many gistokhimik consider this coloring specific to a glycogen, recommending it to use at a research of the fabrics rich with the mucopolysaccharides, mukoproteida, glycoproteins which are painted equally with a glycogen at other techniques. Processing of control cuts a diastase is necessary. It is supposed that the main reagent of dye of Best is carminic to - that, selectivity a cut to paint a glycogen depends on a number of physical and stereovolumetric factors.
